69 Immunophenotyping of TCR and BCR clonotypes

Background T-cell receptor (TCR) and B-cell (BCR) repertoire profiling holds great potential for understanding disease mechanisms the development of new therapeutics in infectious diseases, autoimmunity immuno-oncology. However, this could be greatly improved by combining information about clonotypes with immuno-phenotypes T B cells. Methods To facilitate these studies, we developed a novel technology combined all human TCR BCR variable regions phenotypic characterization immune cells bulk at single-cell level PBMC cell fraction samples. The TCR/BCR Immunophenotyping method involves multiplex RT-PCR amplification sequencing CDR3 genes set most informative T- phenotyping genes. Bioinformatics analysis NGS data allows clonotypes, identification major subtypes their activation status. Results Data will presented showing how clonotype targeted expression can applied large-scale discovery typing biomarkers several immune-responsive model systems. Conclusions Preliminary studies demonstrate assay has unparalleled throughput, sensitivity, cost-effectiveness high-throughput immunity biomarker applications.
